Investigation of Wheat ERF Family Revealed Novel Genes Involved in Powdery Mildew Responses

摘要: Powdery mildew is a global devastating disease that usually leads to great yield losses in wheat production. As a large transcription factor family in the plant kingdom, members of the Ethylene Responsive Factor (ERF) family have received widespread attention for their roles in regulating defense response. Mining genes for resistance breeding is considered as an effective way to control wheat powdery mildew. ERFs involved in powdery mildew response are largely unknown in wheat. Therefore, identification of ERFs in response to powdery mildew will help to understand the mechanism of defense response and provide candidate genes for wheat disease resistance breeding. A series of bioinformatics approaches were employed to isolate the APETALA2 (AP2)/ERF superfamily and the ERF family from the genome of bread wheat Chinese Spring (CS). Transcriptome data were used to screen ERFs involved in powdery mildew response. Subsequently, quantitative real-time PCR (qRT-PCR) was performed to investigate the expression patterns of these screened ERFs in vivo. In this study, the AP2/ERF superfamily was isolated from the genome of bread wheat CS, and a total of 96 ERF family members, including 229 ERF sequences in the A, B and D genomes, were identified according to the clustering results and structural characteristics. Based on the analysis of clustering and transcriptome data, 13 pathogen-inducible ERFs with highly sequences similarity to the reported disease resistance-related ERF genes were further screened. Expression patterns of the 13 selected ERFs in susceptible and resistant wheat materials after Blumeria graminis f. sp. tritici (Bgt) inoculation were analyzed in vivo. Collectively, our results showed that after Bgt infection, 13 screened ERFs displayed different transcript kinetics, indicating different roles of these ERFs in powdery mildew resistance. The discovered ERFs could be used as candidate genes for wheat disease resistance breeding.
