摘要: By analyzing residual hemicellulose after enzymatic hydrolysis of corn husks, we identified previously unreported side-chain architectures in glucuronoarabinoxylan and xyloglucan of corn husk hemicellulose, including alpha-galactosyl-mannose and alpha-xylosyl-arabinose linkages, which shield glycosidic bonds from enzymatic cleavage. These discoveries unveil novel structural complexes in hemicellulose and broaden the understanding of hemicellulose recalcitrance beyond existing literature. Further, we designed a tailored enzyme cocktail by heterologously expressing three targeted hemicellulases (GH51 alpha-L-arabinofuranosidase, GH27 alpha-D-galactosidase, and GH31 alpha-D-xylosidase) based on hemicellulose structural information, and synergistic integration of these enzymes with Penicillium oxalicum cellulase via response surface optimization. This enzyme system achieved unprecedented yields of 81.82 % glucose and 54.89 % arabinose through one-step hydrolysis without chemical pretreatment, surpassing previous benchmarks by over 30 %. This study advances the understanding of hemicellulose's intricate structural complexity and enhances insights into multi-enzyme synergies during biomass degradation, offering a pivotal framework for developing pretreatment-free, eco-friendly enzymatic hydrolysis processes and enabling high-value utilization of corn husks.
