Identification of ABCG transporter genes associated with multi-insecticide resistance in Plutella xylostella (L.)

摘要: Plutella xylostella (L.) is the most severe pest that feeds on cruciferous plants. With the widespread and long-term use of insecticides, P. xylostella has developed resistance to almost all kinds of insecticides, and multi-insecticide resistant populations are frequently reported. However, the mechanisms of multi-resistance in P. xylostella remain unknown. ATP-binding cassette (ABC) transporters, as the largest transport family in organisms, are involved in the detoxification of insecticides. In this study, the roles of a total of fifteen ABC genes belonging to the ABCG subfamily of P. xylostella in multi-resistance were investigated. The results demonstrated that the relative expression of eleven ABCG genes including ABCG1, ABCG2, ABCG5, ABCG6, ABCG7 ABCG9, ABCG10, ABCG11, ABCG12, ABCG13, and ABCG14 could be induced by at least one of the eleven tested insecticides in P. xylostella. The combined results derived from RNA interference and transgenic fruit fly system showed that ABCG2 was involved in the resistance to beta-cypermethrin and indoxacarb, and ABCG10 was associated with the resistance to beta-cypermethrin. These findings are helpful for further understanding the mechanism of multi-insecticide resistance in P. xylostella and other insect pests.
